Preparation of Samples for Aflatoxin Testing
Permitted levels of this
toxin are very low - usually 5 ppb or less. Analysis was performed to ensure
that these restrictions were not violated, and the sampling and testing process
is regulated by law in most countries. However, test correction has not been
combined with any standard method.
A study conducted by the
authorities has found that the preparation of samples by scurrying offers
significant benefits in addition to drying.
The process
As mentioned above, in the absence of a standard procedure, there are many variations in the effectiveness of the preparation (or extraction) process, depending on the method and type of equipment used. The volumes used for sample preparation will also vary depending on the product being tested.
65 - 90 lbs samples are rare.
The usual procedure would be as follows:
The problem
Sample preparation is subject to several issues:
Effective reduction of particle size and sample homogenization before extraction is essential for accurate and repetitive results.
Other methods of dispersing solid objects, especially on a small scale in the laboratory are difficult to verify and can lead to discrepancies between clusters.
Milling machines may be difficult to clean, exacerbating potential pollution/hygiene issues.
The output phase is subject to a lot of variabilities due to the methods used, as described in the previous diagram. Once again it is difficult to confirm.
Dry digestion can cause severe heat loss.
The viscosity varies greatly from liquid to liquid.
Solution
Experiments have found that the preparation of samples by brushing using tools such as the Wahal Engineer high shear mixer offers significant advantages over dry grinding:
Very low CV (coefficient of variation) is available.
The particle size can be reached.
Better homogenization of samples.
Consistent effects throughout the viscosity range.
Items of various sizes can be processed.
This efficiency and consistency between clusters are based on
Wahal Engineer's head mixing function, which works with the principle of the rotor-stator as follows:
Stage 1 Rotating a large rotary speed inside the working head creates a strong absorption of fluid and firms up
to the working head. Stage 2 The Centrifugal force drives the material toward the surrounding surface of the work surface where
they are under the grinding action where it is allowed between the tips of the rotor blades and the
inner wall of the stator. Stage 3 The solids are extracted by the stator and produced back in the body of the mixture. With a short
cycle of mixing all the contents of the vessel beyond the subject matter, it gradually reduces the
particle size to form a perfectly consistent product.
Benefits
Reduced cleaning requirements.
Processing times are greatly reduced.
Laboratory units allow for accurate prediction of the performance of large units when enlarged.
